A simple, rapid, accurate, precise and economic RP-HPLC method was developed for the estimation of aprepitant in bulk and pharmaceutical dosage forms using C18 column (250X 4.6 mm i.d; 5µm) in isocratic mode, with mobile phase comprising of methanol and water in the ratio of 90:10 (v/v). The flow rate was maintained 1 ml/min and the detection was carried out at 220 nm with variable wavelength detector. The retention time (tr) of aprepitant and the rimonabant hydrochloride (IS) was found to be 4.383 and 5.783 min, respectively. The method has permitted linear response in the concentration range of 1- 100 µg/ml and the limit of detection and limit of quantification was found to be 0.130 and 0.395, respectively. The percentage recovery was found to be 99.56 to 101.5 and the % RSD of system precision and method precision was found to be 1.20 and 0.561, respectively. Rimonabant hydrochloride was used as an internal standard in the present study. The method was validated as per ICH guidelines for various parameters such as accuracy, precision, ruggedness and robustness.
